Hope for BRAFers

I'm happy to report that after multiple attempts with multiple standards treatments, I have a found a trial that is promising for folks with the BRAF mutation. It does seem that some of us respond to agents such as folfox of folfiri, but I don't seem to one of them. The triplet therapy associated with the Beacon trial worked like a charm on me for three months, then became emphatically resistant. this new protocol aims to address the pathways that cause resistance. If anyone is interested, I can try to explain. At this point, however, I just hopeful I get into the trial soon -- first on the waiting list for the cohort starting immediately. If that doesn't work, I should be in the second cohort in 2 months.

I would be interested in hearing from folks with this mutation. What has worked for you?

1/8/19 DS Stage 4 with Liver Mets; Successful Colon Resect
2/18/19 Started Folfox -- CEA 70
5/8/19 - BRAF mutation -- switch to Triplet Therapy: Encorafenib, Binimetinib, Cetximab
6/13/19 - CEA dropped from 214 to 22
8/29 - CEA jump to 30-- scans reveal liver spread, though still confined. triplet therapy abandoned; some concern about PIC3 mutation interfering with BRAF treatment
9/1 - 10/4 -- no treatment
10/4 -- folfoxfiri to stem further progression; pump placement in January (hopefully)

Hi, it is hard to do like oxaliplatin? My dad is trying to do beacon regimen?

I did not find the original triplet therapy difficult -- certainly not compared to oxalyplatin. The Erbitux does produce a rash, but that is cosmetic. And I did find myself nauseated a bit more often, frequently without warning.

Is your mutation v600e? Do you know if the triplet therapy can be given thru a gj/feeding tube?

Is triplet regimen easier than your other regimen? How do you rank the regimen from easy to hardest?

Hi there,

My husband has colon cancer stage IV and has the BRAF mutation. He’s been on Folfox regimen. He had five rounds of chemo and a scan that showed that tumors in the liver were shrinking. His doctor from MSK wants to continue with folfox and wait to use the triple therapy for BRAF when needed. We are going to get a second opinion with a doctors from Cornell in NYC. What is your opinion

Hi, do you have more info on this trial? I'd love to know more about it - where it is, etc...
Dx June 2020, stage IV, w liver mets in both lobes. M, age 50. Right-sided colon tumor. CEA 120.
BRAF+ TMB 5% MSS TDL1-1%
July 2020 - Present: 55 cycles chemo (All the various 5-FU regimens)
December 2020 - February 2021 Y90 Radioembolization, Chemoembolization x2
