Tell me any Genome testing

Postby Spazzyjanet » Sun Oct 21, 2018 3:46 pm

Has anyone on this board done genome testing for compatibility with Avastin and/ or Cetuximab and come up with a negative for both?

I am still waiting on the results as tested on my tumor. However, I have my raw data file from 23 and me, and ran it through a membership with selfdecode.com (which cross-references your file with studies) and got scary results against both of them.

Specifically, regarding the IGF1 Gene,

I have rs 6220, genotype AA, which is associated with "shorter overall survival in Metastatic CRC treated with Avastin (Bevacizumab)

I have rs 2946834, GG linked to IGF1, which is associated "with resistance to the drug Cetuximab"

So, question #1- Is this the same thing as genomic testing of the tumor itself or am I way out in left field?

If it is the same, and these drugs can't be added to my FOLFOX +Oxi routine to fight my liver Mets, how far does this set me back? With 6 Mets, (3 left, 3 right), surgery is not yet off the table. Can that, and the standard regime save me?

KRAS G12V (resistant to Cetuximab)
CEA levels normal at diagnosis

Genetic testing is testing against you. Genomic testing is testing the tumor.

Genomic testing typically looks at several hundred known cancer mutations and reports back on which one(s) are present in the tumor.

From your genetic testing results: it looks like there is known information on Avastin with the indicated genetic marker. There may or may not be issues with the Genomic Testing as well.

I guess I will talk to my oncologist about it tomorrow, and hope that his eyes don't roll too far up into his head like they have in the past when I reference my genetic testing.

We'll also see what his tests say. Honestly, with liver mets I am terrified about my prognosis if we can't use Avastin.

Hi!
I can recommend you HAI pump. I used it myself with 10+ liver mets. The largest was 7.5cm. I had 3 liver resections to cut them out but I am here, doing good, off chemo. I went to MSKCC seeking Dr. Kemeny's help.
I also used FOLFIRINOX/FOLFOXIRI at first line chemo.
At that time (2015) there were studies that FOLFOXIRI+ Avastin gave the statistically longer survival and more chance of resection. Dr. Kemeny opted for FOLFIRINOX for me. She dropped Avastin since she wanted me to get resections fast.
I don't read that much now so there must be more newer info than what I know.
I did CARIS genomic testing. I had another test at MSKCC and once more at Mt Sinai for fruit fly study. CARIS testing of my tumor proved accurate.
My approach as a MSS patient was to attack the cancer with chemo fast and massively and go to resection. This old fashioned approach seemed to work in my case. At least until now.
Good luck!
mom of now 14 years old twins, dx @ 40 in 6/2015 with upper rectal cancer, 10+ liver mets, CEA 140
chemo: 8/2015 - 10/2016 - 4xFOLFIRINOX, 2xFOLFOX, 8xFOLFIRI, 10x5FU, HAI pump -12xFUDR
4 surgeries, complete pathological response
CEA<2 since 10/2015
NED since May 2016

I praise God for every day with my family!
