Good news from trial NCT02688712, for Stage II rectal cancer receiving first treatment, to illustrate why and how clinical trials shouldn't be considered something to try after everything failed.

* Chemoradiation+ Xeloda or 5-Fu + IMMUNOTHERAPY (TGFβ Type I Receptor Inhibitor LY2157299)

* Only in Providence, Oregon.

ExIST Study of LY2157299 (Galunisertib) in Rectal Cancer

''The first three patients to participate in the clinical trial have shown a “dramatic response,” according to Dr. Young. One patient’s tumor completely disappeared, while the two other patients had more than a 75 percent decrease in tumor size, with equally impressive responses in the lymph nodes. ''

''The trial is open to patients with Stage 2 and higher rectal cancer who are slated to undergo the standard treatment of radiation and chemotherapy prior to surgery. The clinical trial adds an additional component to the pre-surgery radiation/chemotherapy regimen – immunotherapy. It is administered in the form of daily pills for two weeks prior to radiation and chemotherapy, and then during that treatment as well.''

That is excellent news, in time maybe will get to the point where rectal cancer will be treated only chemically with radiation.
55 year at the time of diagnosis, male
Diagnosed with T1,T2 N0 M0 rectal cancer
Total neoadjuvant therapy or TNT (chemoradiation followed by systemic chemotherapy)
Negative since Feb. '17
No surgery
Watch&Wait approach 8)
I don’t come much to the forum , so if this is not updated it means I remain negative!

I'm for "no radiation" !!!

My GI surgeon at MSK did a trial involving a chemo-only approach to rectal cancer. I think adding immunotherapy to the mix might be just what's needed to increase the efficacy.

I didn't have radiation but only because I was a Stage 4B at diagnosis. There was no plan for surgery of any kind, just chemo for life. When MSK decided to take a chance on me, they went straight to surgery, no radiation whatsoever. At that point, I'd been on chemo for nearly 2 years and had a significant response to it. My GI surgeon said it's much easier to operate on rectal cancer patients who haven't had radiation because it thins the tissue and makes it brittle. Also, patients who don't have radiation have a better outcome in terms of bowel frequency, urgency, etc. than radiation patients. Getting away from burning the delicate tissue in that area would be a godsend.

I sure wish this had been around when I was going through all of this. Radiation was the worse for me. The very reason why I have a permanent colostomy bag today.

Y-90 radioembolization on 9/17/18, liver enzyymes have dropped. 10 Radiation treatments to primary tumor completed too. CT scan Nov to assess overall situation...
