I am still on ADAPT and CT last week shows still NED since last surgery in August 2014.

I have a good quality of life and so will probably carry on with this. I take cimetedine as well.

June 20 CT shows nodule, bronchoscopy confirms is scar tissue, still NED
Dec 20 stopping Xeloda continue celebrex, cimetedine
Aug 21,March 23 scans show still NED
March 2023 CURED - discharged from Oncology, no more scans or follow up

I really don’t have much side effects. The only thing that I get is a stool that somewhat burns, almost like a virus, when on the Xeloda. Other than that, I feel pretty normal.

It was not part of a trial but I had my local onc call Dr Lin and get the protocol for me to follow and my onc at MDA neither approved nor disapproved of me adding the Celebrex. He finds it intriguing that I have no HFS and I keep telling him it is bc of the Celebrex!!

I was on the ADAPT protocol for three years. 2000 X/ 400 C. I never got HFS either- I never made the connection with the Celebrex! That’s very interesting.
I went off this past Memorial Day because I’d been NED for some time. I have a scan 11/8.

Thank you Donna and great news on being able to come off of adapt. I wish you NED forever!!! How did you get approval for it? Did you ask you oncologist or did they just put you on it? I'm taking 2000mg of xeloda a day but just not convinced it's doing enough to keep me NED.

My daughter has been stable on this protocol for 2.5 years, not as part of the trial though. I had run into this trial diring my research to help my daughter. I was waiting for the right time to approach her onc with this so when he wanted to give her a chemo holiday in Jan ‘18, she was afraid. Thats when I suggested Adapt. He was positive towards it but said he’d like to add avastin. She’s done very well so far. Mild/moderate hand/foot but feels good and is living her regular life... a great blessing. I don’t understand what happened to this trial. I’ve run into numbers of people still on it or implementing it on their own. I’ve heard that Dr Lin is back at Fred Hutch in Seattle. I also heard there might be a similar trial being designed but I don’t know....anybody?

Thanks to everyone in this thread, I am writing this down and hope to bring it up to my oncologist when I finally get to see him (or her)

Seems like Celecoxib increases heart risk, but I guess for most of us this is an acceptable risk.

I've been taking celecoxib since December 2019 for the bad joint pain I've suffered as a side effect of Keytruda. Everything still hurt, but significantly less.

What is it supposed to do as part of the ADAPT protocol? My understanding is that it is a COX2 inhibitor, so helps with inflammation. I've never seen anything about anti-cancer effects.

My wife took celecoxib at 300-400 mg per day, with her daily chemo (and lots of carefully selected supplements), for over five years. Early in our chemo-supplements-celecoxib series, her markers rose but liver panels were good at 200 mg. At 300 mg her markers were stable or slightly depressed and liver markers very slightly affected; at 400 mg her markers improved but her liver panels had some rise. Over time we adjusted mostly UFT-LV loading and supplements for a better therapeutic index. After chemo she continued celecoxib for part of a year. She still does a fair amount of supplements e.g. IVC, big D, vit K2, beta glucans, resveratrol etc.

My wife started celecoxib at 2.8 years out from diagnosis to increase chemo's effect. Along the way she had 2-3 periods of hypercoagulability, that might be cancer, chemo and/or coxib related.
Stop? Damned if you do, damned if you don't - markers can rise and run.

Basically if she ran short on any category, chemo (UFT/5FU-LV), IV vitamin C, supplements, or celecoxib, her marker(s) would rise. So sometimes it became a juggling act to beat down coagulability and cancer markers, mostly by adjusting celecoxib and supplements several ways based on their various properties and case specifics.

She has been chemo free for two years now, after two surgeries and 8 years of chemo and 10 years of supplements. Careful, combined use of celecoxib, oral chemo, and substantial selected supplements worked for us.
watchful, active researcher and caregiver for stage IVb/c CC. surgeries 4/10 sigmoid etc & 5/11 para-aortic LN cluster; 8 yrs immuno-Chemo for mCRC; now no chemo
most of 2010 Life Extension recommendations and possibilities + more, some (much) higher, peaking ~2011-12, taper chemo to almost nothing mid 2018, IV C-->2021. Now supplements

I am so happy for you two that she is off chemo!!! Hopefully forever!!!
Both of you are are such an inspiration!!!
It is a juggling act for sure to get the markers down and keep them down.

I've always been interest in Celebrex and recently ran across various versions of this

I'm not good at reading science...but seems to indicate a benefit to those with a SMAD3 mutation (which I have)
Not sure how common SMAD3 is as its not part of the normal genetic test panel. Might be worth a look for some.

I'm going to ask my Doc to prescribe Celebrex on my next visit.

I have been on Celebrex 400 daily for more than five years. Initially with xeloda for maintenance and then just Celebrex. I have had a good oncological outcome (Stanley Scans for five years off chemo) but did develop rapid onset coronary artery disease and will be having a quadruple bypass this week. Not sure if u will continue the Celebrex

12/2014-4/2015: FOLFOX (8 cycles)
4/2015-6/2015: 28 cycles of chemoradiation with xeloda, SBRT
8/2015: Robotic APR with iliac node dissection; path showed ypT0,ypN0 (complete pathological response).
11/2015 scans clear, CEA 2.1
11/2015 parastomal hernia repair
3/2016 CEA 1.7, scans stable...
6/2020 5 years of normal CEA and stable scans
Now dealing with pyoderma gangrenosum.
Totally disabled due to oxaliplatin induced neuropathy and dysautonomia
