Oncologist told us there are currently 91 open slots in the trial nationwide. He also said that in initial Phase I trials a few years back, the side effects of BBI608 were so harsh he had little hope for the drug. He now feels very positive about the results they are seeing, and said they seem to have found the "sweet spot" for dosage in terms of limiting side effects.

Most recent scan last week showed control/minor shrinkage on Xeloda/Avastin, so we are sticking with that for now and keeping trial in our back pocket. Just thought I'd share for those interested.

Thanks for the info. I had heard the side effects were similar to Folfiri alone and I hope it's true.
I got the trial nurse at one of the centers to take my records finally, so I guess I'm on the road to finding out if I will be in this trial. Fingers crossed.

Hope you hear back soon on the trial Elizabeth. Question for you. Have you been on folfiri since sept 2015 continuously? And it's been keeping you stable all this time?

Yes. I've had 39 treatments over tge past 18 months with only a week break here and there for holidays or vacation. I am stable-ish. My CEA was creeping prior to my microwave ablation but the cause wasn't evident on scans. My CEA is trending down currently. The side effects have changed too. My hair has started to come back and the diarrhea decreased. I'm just more fatigued.

Quick update... scans in July showed minor progression on Xeloda/Avastin. Received word today that my husband has been accepted into NCT02024607 (BBI608, also known as Napabucasin + FOLFIRI arm) at Mayo Clinic. He will take his first dose of the drug this Friday 9/1, and will get his first infusion of FOLFIRI on 9/6. They will scan again after 4 infusions. Fingers crossed that he can tolerate the drug, and that we see some tumor regression.

My husband had two stable scans on the BBI-608 trial with FOLFIRI. At that point, due to some unrelated kidney problems (ureter obstruction caused by radiation) he was unable to receive FOLFIRI for seven weeks and unfortunately the next scan showed progression. However, growth WAS controlled when he was able to have chemo, so if you are considering this trial, please don’t rule it out. Others have had stable disease for up to a year.

Kidney problems have been addressed with nephrostomy tubes. We’re looking at another trial option at Mayo and hope to start in a couple of weeks.
