BRAF and RAS (KRAS or NRAS) mCRC patients. Trial -Update 12-29th

Postby Maia » Sat Nov 19, 2016 11:22 am

BRAF and RAS (KRAS or NRAS) mCRC PATIENTS. Maybe of the interest of those with p53 deficient or mutated too.
A trial (update:) recruiting at Dec 29th 2016. It's not immunotherapy, but irinotecan plus AZD1775 (a WEE1 inhibitor; "a small molecule inhibitor of the tyrosine kinase WEE1 with potential antineoplastic sensitizing activity").

Posting because it's an area of high unmet need.
https://clinicaltrials.gov/ct2/show/NCT02906059

It has been at least a Phase I Study Evaluating it Monotherapy and in Combination With Gemcitabine, Cisplatin, or Carboplatin (Solid Tumors) http://ascopubs.org/doi/abs/10.1200/JCO.2016.67.5991 and a Phase II study with it plus carboplatin in patients with p53 mutated ovarian cancer refractory or resistant (< 3 months) to standard first line therapy http://meetinglibrary.asco.org/content/149374-156 More: https://ncats.nih.gov/files/AZD1775.pdf

this is where I put my hand up
I'm new here and new to the world of CRC . I sort of understand that braf, ras, kras is "versions" of a cancer or particular mutations of it....
but whatsitmean??? I mean I know I have colon cancer that started in my sigmoid colon but no one has told me what version I HAVE tho that might be because I'm just starting my "race" here and haven't even had my first Chemo (Nov 22nd) OR it could be that I'm in Canada and that test result hasn't reached me in my Igloo yet  Canada post is a little slow sometimes

Other trial for BRAF mutated CRC:

NCT02906059
Study of Encorafenib + Cetuximab Plus or Minus Binimetinib vs. Irinotecan/Cetuximab or Infusional 5-Fluorouracil (5-FU)/Folinic Acid (FA)/Irinotecan (FOLFIRI)/Cetuximab With a Safety Lead-in of Encorafenib + Binimetinib + Cetuximab in Patients With BRAF V600E-mutant Metastatic Colorectal Cancer (BEACON CRC)

Many locations.
Even if not applying for the trial, to keep in mind combination that are being trialed for BRAF + mCRC

"Phase 3 study to evaluate encorafenib + cetuximab plus or minus binimetinib versus Investigator's choice of either irinotecan/cetuximab or FOLFIRI/cetuximab, as controls, in patients with BRAFV600E mCRC whose disease has progressed after 1 or 2 prior regimens in the metastatic setting."

I think different chemos work better with different mutations. Or KRAS works with immunotherapies. Hopefully someone else answers you. I'm probably wrong :D
