MSS-CRC: PD1/NKTR-214 Combo Trial Announcement

A new MSS-CRC PD-1 combo trial announced = "NKTR-214". No NCT# yet just press release. In preclinical CRC models they saw activity of single agent - but as usual, my guess (but just a guess) is that clinically - a combo with PD1 is more likely to be successful.

As an aside: As I completely expected, since the Cobi/Atezo "Proof of concept" we are now going to be overrun with new MSS-CRC combo trials to choose from over the next 12 months, today is just the latest example. On one level a good thing (many choices) but also an issue since how do you choose? But still a better situation than a year ago when MSS-CRC immunotherapy trial options were tough to find and doing any immunotherapy trial for MSS-CRC was not easy since demand outstripped supply of slots...

You're right: there is an abundance of "me too" PD-1-combo trials coming out. Better too many than too few though. I think everyone in the checkpoint inhibitor space is busy doing deals with smaller innovative biotech companies, looking for a breakthrough treatment. There is plenty of opportunity for serendipity now, but as the field matures, this could give way to a more systematic approach. Fingers crossed for all MSS-CRC patients! :D

Tom: Strangely, UNC was in the Phase 1b testing for Cobi/Atezo but is not a site for the Phase III testing. Any thoughts why this might be? I wonder how many "adverse events" or toxicities? I am meeting with a Duke oncologist for a consult next week, I will pick his brain about this. Sorry if this deviates a bit from the topic...

Impossible to guess why UNC was not in the Phase 3 list - probably many possibilities behind it.
I have heard mixed things on the toxicity of the combo - would be interesting to hear your oncologist's take.
