Ieowi,
I lately learnt that the mutation your dad and I hold FBXW7 is highly immunogenic. He is also MSI low. Considering all this I think there is a good chance he might respond to immunotherapy meds.

"However, mutations in FBXW7 were associated with the absence of metastases and were seen in M0 patients across cohorts. This correlated with increased expression of T-cell proliferation and antigen-presenting functions. Dr Fox said that mutated FBXW7 increases inflammatory signaling, thereby boosting the immune response."

All the info about significance of this mutation as triggering immune response are relatively new, hard to find.

Very interesting about the Diclofenac. I wonder if the mechanism is similar to that of celecoxib (Celebrex) given that both are arthritis medications. Dr. Lin's studies seemed to indicate that those with the PIK3CA mutation were better responders to his ADAPT protocol, which is celecoxib and Xeloda. ADAPT is targeted at those who are NED, but I wonder if the celecoxib could at least help to slow growth in those with active disease? Might be worth asking your doctor if you haven't already.

Life Extension has advocated similar COX inhibitors for 20 years, as well as aspirin for CRC, although it is still unclear to me about precise differences on the different COX2 inhibitors and possible combinations. We've use 81 mg aspirin and 300-400 mg celecoxib combined for a couple of years. LEF's Lodine recommendation was closest to diclofenac.

"In 1996, Life Extension recommended that most cancer patients take a COX-2 inhibiting drug because of solid evidence that cancer cells use the COX-2 enzyme to sustain their rapid division. In 1996, Americans had to import a COX-2 inhibitor named nimesulid from other countries because this class of drug was not widely available in the United States."

"Experiments in laboratory animals suggest that drugs such as Celebrex could help cure cancer, especially if combined with chemotherapy or radiation (Hsueh et al. 1999; Pyo et al. 2001; Swamy et al. 2002). There are 100 separate cancer studies involving COX-2 inhibitors going on worldwide at this time."

"Doctors are predicting that COX-2 inhibiting drugs may become standard therapy in 5-10 years. There was adequate evidence in 1996, however, to recommend COX-2 inhibiting drugs available to cancer patients."
watchful, active researcher and caregiver for stage IVb/c CC. surgeries 4/10 sigmoid etc & 5/11 para-aortic LN cluster; 8 yrs immuno-Chemo for mCRC; now no chemo
most of 2010 Life Extension recommendations and possibilities + more, some (much) higher, peaking ~2011-12, taper chemo to almost nothing mid 2018, IV C-->2021. Now supplements
