Postby vilca11 » Sat Jun 13, 2015 12:11 pm

Yesterday got from Dr. Nilo Azad/Hopkins ..... did we talk about this study? It is not new, opened in 2013, I wonder why she tells me about it now.... may be they have good results in cohort 1 and 2 and are proceeding to cohort 3 and 4... Will find out and report. Opinions from trial gurus??? Hugs, vilca

Here is some history about the CVAX technology:

https://en.wiki2.org/wiki/GVAX

Here is the current owners of the technologies web site:

http://www.aduro.com/about/

Its hard to say where this all stands. Looks like Aduro is working on this for Prostate, maybe Ovarian cancers. Early attempts though showing promise I don't think were very successful. They are now trying it in combinations with other agents including PD-1 inhibitors. Your friends at John Hopkins might be able to tell you more about heir particular trial. Its at least 2 and a half years out before completion from the looks of things.

Here are information about the two Biotechs I think hold a lot of promise.

https://en.wikipedia.org/wiki/Northwest_Biotherapeutics A DCVax type therapy, and

http://www.oncolyticsbiotech.com/

The later is running quite a number of ongoing clinical trials.

I haven't personally heard buzz about this particular mCRC trial - doesn't mean it isn't working - just that I have not personally heard any promising information on it.

Where GVAX is getting a lot of buzz and clinical success is via cotherapies, in other cancer types:
1.) CRS-207 + GVAX won a "Breakthrough Therapy" designation from the FDA for pancreatic cancer and is looking like an immunotherapy step forward against that dreadful ^ aggressive disease http://investors.aduro.com/phoenix.zhtm ... ID=2025657
2.) Multiple planned trials with checkpoint inhibitors e.g. http://www.fiercevaccines.com/story/adu ... 2015-03-03
3.) Other indications as GrouseMan mentioned.

So... unfortunately not a lot of guidance on how to judge the potential merits of the CRC trial. Aduro may not be focusing on CRC either for business or scientific reasons - really no way of knowing for sure from the outside...

In a general sense, I believe that similar to viral therapies, vaccine therapies which have languished for a few decades with minimal clinical efficacy will be resurrected by the current & coming wave of immunotherapies, kicked off with the checkpoint inhibitors. One of my favorite preclinical papers this year showed the power of a combo of: 1.)vaccine 2.) Checkpoint inhibitor (adaptive immune system) 3.) innate immune system activator - totally demolished tumors including hard to treat ones like CRC. That is the type of immunotherapy wave coming rapidly down the pipe towards the clinic........

Please let us know if you "hear anything" from the clinical trial team!!

Thank you so very much for the answers and your time, guys! It is so wonderful that we have you on the board to steer us towards the most promising ones and explain us all the details in lay language. And, besides, we have a chance, thanks to both of you, Maia and Joe777 we have info about everything new faster than many others. So, just lots of gratitude goes your way from everyone, I know....
